Detection of virus-specific glycoproteins within herpes simplex virus transformed cells

Abstract
Mammalian cells transformed by herpes simplex viruses type 1 and type 2 (HSV-1 and HSV-2) were examined to determine which of the major virus-specific glycoproteins are expressed on the membranes of the HSV-transformed cells. The major envelope glycoprotein regions of HSV-l and HSV-2, VPl23 and VP119, respectively, were purified by preparative SDS-polyacrylamide gel electrophoresis and were separated by SDS-hydroxylapatite chromatography. Various glycoprotein components were resolved: in the HSV-l system, components were designated 1AB and 1C; in the HSV-2 system, components were designated 2AB, 2X, and 2C. An antiserum to each of these highly purified glycoproteins was used in membrane immunofluorescence, radioimmunoprecipitation, and competition radioimmunoassay studies with HSV-2 transformed rat, human, and hamster embryo fibroblasts and HSV-1 transformed hamster embryo fibroblasts. Analysis of the HSV-2 transformed rat and human cells showed that antisera to the 2AB and 2X glycoproteins were reactive with these transformed cells; however, no significant reactivity was observed with the antiserum to the 2C glycoprotein. HSV-2 and HSV-1 transformed hamster embryo fibroblasts were also examined for HSV glycoprotein expression and were found to express the 2AB, 2X, 2C and 1AB and 1C glycoproteins, respectively. These findings are correlated with recent physical mapping studies of the portions of the HSV genome present within the transformed cell.
